Slow Growth In Vitro Conservation Studies in Garlic

The effect of different osmo-regulator concentration and their combinations, over in vitro slow conservation of garlic at ambient temperature against period and other morphological growth parameters was investigated. Protocol was mainly developed to achieve conservation of genetic resources free from disease and pest and to reduce cost incurred towards maintenance under field and cryo-conditions. Garlic, being exclusively clonally propagated bulbous crop, major occurrence of quality seed deterioration happened due to seed borne pest and disease. In present studies efficient slow growth conservation protocol was developed at an ambient temperature. Plantlets developed from meristem, then after establishment in medium they were individually placed in test tube with MS medium supplemented with 12different combinations of sucrose (0.1M, 0.2M, 0.3M, 0.4M) sorbitol (0.1M, 0.2M, 0.3M, 0.4M) and combination of sucrose and sorbitoleach with concentration of 0.1M, 0.2M, 0.3M, 0.4M along with control. Culture room maintained at 25 ± 2ºC temperature, light intensity for 16:8 h photoperiod with 50-80 % relative humidity. After fifth months of storage, treatment combination MS medium supplemented with 0.2M sucrose + 0.2M sorbitol demonstrated positive retarded growth at normal temperature coupled with 70 % survival. However, in overall experimentation cultures survived across genotypes ranged from 20 -70 %. Plantlets cultured in this medium are free of contamination. As far as we know there are no published reports on in vitro storage of garlic at ambient condition.
